Can Ancient DNA Test Cain’s Arrival?
A study that cannot yet be run, and an account of what is missing

This site proposes, with modest confidence, that Cain crossed into our world near the Zagros foothills, in what is now western Iran, around 5000 BC. If a genetically distinct person entered an already-peopled region and left descendants, that should in principle leave a trace: ancestry appearing in western Iran after that date which was not there before.
This page reports what happened when I asked whether that test can be run with the ancient DNA currently published. It cannot, not because the answer came back negative, but because the data thins out precisely where it is needed. What follows is the accounting.
Where it stands
The test is possible in principle and badly underpowered in practice. Individuals living after the proposed date have been sequenced, and could in principle carry the ancestry of an earlier arrival. But the nearest published baseline before that date is a single individual some eight hundred years older, from a different part of the region. Any difference found between them has several ordinary explanations before it reaches this one.
What has been sequenced
Drawn from the Allen Ancient DNA Resource, a large public database of published ancient genomes, version 66, the published record for western Iran around the relevant centuries is this:

Khuzestan, the lowland plain named in this site’s proposal, has no published ancient human genomes at all. Neither does Fars. The number is not merely small; it is zero.
Why the gap matters, and why it is not fatal
The proposed date falls inside the empty band. That sounds worse than it is: the arrival itself does not need to be sampled. If a line established itself and persisted, its descendants living around 4800 to 4400 BC would carry its ancestry, diluted but present, and several such individuals have been sequenced.
The difficulty is on the other side. Detecting an arrival means knowing what the population looked like beforehand, and the nearest published baseline is one person, eight centuries earlier, from a valley some distance away. Eight hundred years is long enough for ordinary movement of people to change a population considerably. A difference between that baseline and the later group would be genuinely ambiguous, and the ambiguity would not be resolvable with the samples in hand.
Two things decay at different rates here, and it is worth separating them. Ancestry dilutes by half in each generation of intermarriage but remains detectable a long way down. A distinctive lifespan, the kind of unusual longevity this site attributes to the line from the ancient world, would fade faster if it existed, because it requires most of the inherited material rather than a trace of it. A future result showing the ancestry and not the lifespan would therefore be consistent rather than contradictory.
Where the unsequenced material is
The gap is in the sampling, not in the ground. Excavated skeletal material from this period exists; almost none of it has been sequenced. Establishing where it is held turned out to be harder than expected, and the result is mixed.
- Tepe Sialk: thirty-nine skulls survive in the anthropological collection of the Musée de l’Homme in Paris, of which eleven are dated to the late fifth or early fourth millennium BC. The postcranial remains were lost. None has been sequenced. This is the best-documented unsequenced material inside the window, though Sialk lies on the central plateau rather than in the region this site proposes.
- Ganj Dareh: human remains are held at Simon Fraser University in British Columbia, with a minimum of 116 individuals documented. The site is far older than the window, around 8000 BC.
- Tepe Abdul Hosein: twelve individuals, held at the National Museum of Iran. Also older than the window.
- Chogha Mish, in Khuzestan, has a published skull dated roughly 4500–4000 BC, the right region and the right period. Where it is now curated could not be established.
- Seh Gabi, Godin Tepe, Tepe Giyan, Tepe Guran and Chogha Sefid: the current location of the human remains could not be established from any accessible source. This is a gap in the published record rather than in the search.
That last point deserves stating plainly, because it was the most surprising finding of the audit. For several major mid-twentieth-century excavations, it does not appear to be publicly recorded where the excavated human remains ended up.
What would make the test decisive
- Sequenced individuals from western Iran between roughly 5600 and 4800 BC: at present there are none, and even a handful would convert a one-person baseline into a population.
- Material from Khuzestan and the Zagros foothills specifically, rather than the plateau or the north-west.
- Enough individuals on each side for the comparison to mean anything. The equivalent calculation for the Cudi Dağı study suggested around sixteen per side to detect a ten per cent contribution from a highly distinctive source; a smaller or less distinctive contribution would need far more.
Limits of interpretation
No genetic result could identify Cain or any other named person. A distinctive ancestry component cannot recover a name, a language or a reason for travelling, and it cannot distinguish an extraordinary arrival from an ordinary one without independent evidence. Were an unexplained component ever to appear in this region and period, the first response would properly be to seek more samples and better regional models, not to reach for a text.
The present conclusion is narrower, and is a statement about data rather than about history: the published record for western Iran is too thin, and too unevenly spaced around 5000 BC, to test the proposal either way.
